It looks like Joker 2 is confirmed with Joaquin Phoenix returning as the villain, but the rumor is Lady Gaga is in early talks to play Harley Quinn.
Sources are also saying the movie may also be a musical. Which I don't know how I feel about that, but either way I can see Lady Gaga playing Harley.
This Harley Quinn would be in a separate DC universe than Margot Robbie's Harly Quinn character, but so far it's all "being kept under wraps."
